The Series S-3 was the only model offered by DeSoto for 1937. Two wheelbase sizes were available including a 116-inch platform and a longer 133-inch version. Several body styles were offered with the most popular being a touring sedan which saw nearly 52,000 examples sold. The second most popular was the Touring Brougham which had 11,660 sales. Power was from an L-head six-cylinder unit which had a 228.1 cubic-inch displacement and offered 93 horsepower. A synchromesh three-speed manual transmission, hydraulic brakes, and steel disc wheels were standard.

The S-3 styling featured a grille with horizontal bars divided by a vertical center panel with matching body color. The DeSoto name was spelled out by vertically stacked chrome letters on the center panel. In the front were a wide bumper and bullet-shaped headlamps. The all-steel body construction had 14 rubber float body mountings and the new hood had hinges at the cowl.

The S-3 featured 'safety-styled' interiors. The speedometer rested in front of the steering column, and the instrument panel knobs were recessed. The gauges were flush-mounted.

Standard features included bumper guards, spare tire, and safety glass. The convertibles came standard with door armrests, dual wipers, taillights, sun visors, and horns.
